After 25 years, thecause ofarare genetic neurological disease hasfinally been discovered
Spinocerebellar ataxia type 4 (SCA4) isasporadic syndrome. Symptoms include muscle weakness, difficulty coordinating body movements, which causes aparticularly jerky andunsteady gait, anddifficulty speaking. Thedisease haslong been known tohave agenetic cause. Asearly as1996, alink tochromosome 16q wasfound, but themutation causing SCA4 remained unknown until now. Using thelong single-stranded whole genome sequencing (LR-GS) method, which searched 6 495 genome sequence datasets, scientists finally discovered that amutation intheZFHX3 gene wasresponsible forthedisease.

Our genotype can influence thelikelihood ofcontracting tick-borne encephalitis
Thelocation ofthetick does not determine whether someone gets encephalitis. It's different with genes. There isaslight confusion intheDNA that contributes toanincreased risk ofinfection, but also ahigher likelihood that thecourse of"tick-borne" disease will besevere.
A general condition, anencounter with aninfected tick, or theability ofthevirus tocause disease. These areexamples ofwhat influences whether someone gets tick-borne encephalitis. It also includes our genetic makeup. European scientists, including those intheCzech Republic, have found anddocumented achange inDNA that contributes totheoutbreak ofthedisease.
There isone particular DNA substitution that, once you have it, predisposes you tobemore likely tobesusceptible totick-borne encephalitis. Moreover, if you doget it, it ismore likely tobemore severe, says virologist Daniel Růžek, whoworks at theBiological Centre oftheAcademy ofSciences, Masaryk University or theResearch Institute ofVeterinary Medicine.

What ispredictive genetics
It isarelatively young, but rapidly developing medical field, which wascreated onthebasis ofthedecoding ofthehuman genome in2003, aspart oftheHuman Genome Project. Predictive genetics combines genetics with preventive medicine, where theanalysis ofDNA samples can reveal thepredispositions foranumber ofcivilisational diseases andinherited diseases. This allows thenecessary preventive measures tobeset up andtheonset ofdisease tobeprevented intime. Inaddition, thebody's ability tometabolize certain harmful substances or drugs, food intolerances andgene mutations that increase therisk ofcardiovascular or cancer diseases, andmany others can bedetected. Having apredisposition or apredisposition toadisease does not mean that you have thedisease or that you arecertain todevelop it inthefuture. Thedevelopment ofadisease isinfluenced bymany factors. Inaddition tothegenetic code, there arealso external factors, including lifestyle andenvironment, dietary habits, stress andothers. Therefore, anintegral part ofapredictive genetic analysis isaconsultation with aphysician andapersonalized set ofrecommendations onwhat measures totake incase ofapositive result andhow toeffectively prevent therisks ofdeveloping thedisease.
